A comparative study in direct cryopreservative efficacy between Triladly® and EDTA saline glucose 10% glycerol cryopreservative media for human and non-human infective trypanosomes OAK

The efficacy of Triladyl®, a commercial cryomedium for bull semen, in the cryopreservation of both human and animal infective trypanosomes as compared to EDTA Saline Glucose (ESG) 10% glycerol was evaluated in the current study. Cryopreserved Trypanosoma brucei rhodesiense, T. evansi, T. b. brucei and T. congolense were first propagated in irradiated mice. At the peak of parasitemia, parasites were harvested by cardiac puncture and 106,105,104103,102 and 10 dilutions made using whole blood bled from clean mice. These dilutions were divided into two equal portions of 0.5 ml each and cryopreserved in both ESG 10% glycerol and neat Triladly®. Control of gene expression in Trypanosomatidae BJMBR

The study of mechanisms which control gene expression in trypanosomatids has developed at an increasing rate since 1989 when the first successful DNA transfection experiments were reported. Using primarily Trypanosoma brucei as a model, several groups have begun to elucidate the basic control mechanisms and to define the cellular factors involved in mRNA transcription, processing and translation in these parasites. This review focuses on the most recent studies regarding a subset of genes that are expressed differentially during the life cycle of three groups of parasites. Wild animals are exposed to numerous pathogens, including hemoparasites. The Trypanosoma and hemogregarinegroup are frequently reported as parasites in anurans (frogs, tree frogs and toads). The identification of these hemoparasites is usually made through stage observation of their morphology in the peripheral blood of the host. Identification and Characterization of a Trypanosoma congolense 46 kDa Protein as a Candidate Serodiagnostic Antigen OAK

Trypanosoma congolense is a major livestock pathogen in Africa, causing large economic losses with serious effects on animal health. Reliable serodiagnostic tests are therefore urgently needed to control T. congolense infection. In this study, we have identified one T. congolense protein as a new candidate serodiagnostic antigen. The 46.4 kDa protein (TcP46, Gene ID: TcIL3000.0.25950) is expressed 5.36 times higher in metacyclic forms than epimastigote forms. The complete nucleotide sequences of TcP46 contained an open reading frame of 1,218 bp. Southern blot analysis indicated that at least two copies of the TcP46 gene were tandemly-arranged in the T. congolense genome. Stability of Loop-Mediated Isothermal Amplification (LAMP) Reagents and its Amplification Efficiency on Crude Trypanosome DNA Templates OAK

This study evaluated the stability of LAMP reagents when stored at 25C and 37C, and also assessed its detection efficiency on different DNA template preparations. Accordingly, LAMP using reagents stored at 25C and 37C amplified DNA of in vitro cultured T. b. brucei (GUTat 3.1) from day 1 to day 15 of reagent storage. There were no significant differences (P>0.05) in detection sensitivity of LAMP among the reagents stored at 25C, 37C and –20C (recommended storage temperature). Uveíte associada à infecção por Trypanosoma evansi em cães no município de Uruguaiana, RS, Brasil Ciência Rural

Descrevem-se, neste trabalho, as alterações oculares de dois cães naturalmente infectados por Trypanosoma evansi. Os animais apresentaram, ao exame oftalmológico, teste lacrimal de Schirmer I normal, teste de fluoresceína negativo, quemose, hiperemia conjuntival, secreção mucopurulenta, miose, edema de córnea, pressão intraocular diminuída e efeito Tyndall positivo. No esfregaço sanguíneo, foram identificadas formas tripomastigotas, classificadas como pertencentes à espécie T. evansi. Pelos resultados aqui apresentados, concluímos quanto à necessidade de realização de avaliação oftalmológica completa em cães apresentando uveíte, incluindo exames parasitológicos específicos para hemoparasitas.
